Great question. Certain substances including alcohol and some medication, induce the liver to form more enzymes the detoxify the substance. Additionally, the locations were the drugs acts (like the brain) my down regulated the sensitivity to the drug by making less of the proteins that trigger the reaction at the cellular level. There are other mechanism as well that the article I linked to discuss in detail.

I'd like to add: always have at least one, and preferably two alcohol free days a week, to let your liver recover, or you are at much greater risk of cirrhosis of the liver. If you want to minimise tolerance, have one day on, one day off drinking. To reduce hangovers, drink plenty of water before sleeping, and keep a large glass ot it handy, if you need to urinate at night, and drink it then. Men are now advised to not have more than 4 standard drinks /day, and women, 2. Pregnant females, or those planning to be soon, really should abstain; there is NO safe level for the foetus!

The previous answer is good but I just wanted to maybe simplfy it a little. What happens is the liver metabolizes the substance faster thus getting rid of it quicker. Secondly the body makes changes in how its receptors, enzymes and membrane actions respond to the substance. Our body likes to stay in a constant state, this makes it happy:) So it makes changes to stay in that state. The less alcohol, etc that the body's exposed to the quicker the "desired" effects can occur b/c it's not quite as "used" to the substance.
