The meaning is to have faith in Jesus, as demonstrated by the man who was lame (could not walk) and lay on the bed rather than standing on his feet. Once he believed that Jesus could heal his lameness, he was able to walk. He picked up the bed because he did not need it any more as a place to sit.

It is a quote from the Bible. Jesus healed a lame man and told him to take up his bed and walk. The man did.
Now days it is often used by those who feel that the one they are speaking to is not sick, or at least not so sick that he/she could not get out and do something.
