I'm trying to build a tool shed with 2x6 joists and 3/4" plywood on top of it for the floor. I've already built the wall frame. Thanks!

Typically, you finish the subfloor, which includes fastening the 3/4" plywood subfloor, and put your wall framing on top of the plywood. The bottom plate is fastened to the plywood and at several places through the plywood into the 2x6 joist and rim board.

nail it at every stud to your floor, if your worried about wind try using 3 inch screws (coarse thread like sheet rock screws)
