Each type of STD has a separate test. A general practitioner can order the tests if you're a guy, but an GYN is a better source for women.
From blood, they can test for Hepatitis, HIV, and Syphilis. There is a blood test out for herpes simplex 1/2, but not many places are doing it yet. Gonorrhea and Chlamydia are tested using a urtheral or cervical swab, depending on your gender. They might also do a throat or anal swab for gonorrhea, depending on your risk factors. If you have any sores in that region, they can swab the sores and test for herpes.
Pap smears for women will test for HPV and other precursors to cervical cancer. There is no reliable HPV test for men.
The really important part here is that you have to ASK for these specifically, they're not part of a regular exam, even from a gynecologist. Typical blood work will not reveal these things.
