Karate is Japanese and Taekwondo is Korean. In Taekwondo, there are two big federations. International Taekwondo Federation (ITF) and World Taekwondo Federation (WTF). ITF teaches the traditional Taekwondo that focuses on everything from self-defence, sparring, patterns, and breaking. WTF is considered more sport like and does a lot more sparring than the others. I don't know much about Karate but it has a great self-defence system and uses lots of hand techniques (punching). Taekwondo has lots of kicking.
Both martial art's amount of fighting and fitness will depend on the school but both are great ways to lose weight.

This cannot really be defined, since there exist several major systems of tae kwon do and dozens of styles of karate, each with their own particular emphasis and philosophy. Essentially, however, tae kwon do can be seen as a form of karate practiced in Korea.

1) Language. Tae Kwon Do is Korean & Karate is Japanese.
2) Emphasis. TKD emphasizes foot techniques. Okinawan Karate emphasizes hand techniques. Japanese Karate is in the middle.

As for focus on fitness vs fighting, it varies from school to school within each style.
