It depends hugely on which drug. Some drugs have really bad and often permanent physical and mental effects (eg meth, ecstacy), but some drugs are decently safe over the long term (eg ketamine, heroin especially when not injected, and weed).

Social effects are usually more of a concern. The biggest problem is that most people end up having a life that completely revolves around obtaining and doing drugs to the point where they have no friends that don't do drugs, and if they stopped doing drugs they'd have no idea what to do with themselves and how to relate to healthy people. Other issues are money wasted, association with people who might lead then into other criminal activities (prostitution, robbery, etc), the possibility of jail time, and alienation from family members or anyone else who doesn't do drugs.

Also most drugs that make people feel temporarily happier, such as meth and cocaine, have the effect of making the person more depressed in the long term the longer they do them, making the "need" for the drug increase. This mostly resolves after the person has been off drugs for a while, but makes it much harder for people to quit in the first place. With some drugs like ecstacy, this effect is more permanant.
