It is not sufficient to achieve one's goals just to want something really strongly. One has to work all the way from where we start to where we want to arrive. That is, achievement comes through a long series of small successes, and not just through wishful thinking, no matter how much energy we put into wanting. Ambition, nonetheless, is critical to success, in that it defines initially the goals, fuels the efforts, and provides the energy to surpass the failures that will temporarily block any road to success.

Encarta Dictionary's literal definition would be "The quality of being devoted or committed to something in order to do what is necessary to bring about or achieve something expected, desired, or promised, for a strong feeling of wanting to be successful in life and achieve great things." However, what this quote means is that when one creates goals (or ambitions) in life, the only way to realize them (or fulfill them), is to devote (dedicate) the necessary amount of time and energy into seeing them play out. Because only "dedication fulfills the ambition."
