If you need a car when you get there, then drive. Otherwise, the express train is good. I think better than flying because the seats are big like first class seats. Flying is not faster because you still need to get to the airport, arrive early, then go through security, etc. Going by train is very easy from Manhattan. Commuters do it.

If you're willing to pay $200 each way (or about $300 round trip), Amtrak's Acela Express does the trip from NYC to DC in less than 3 hours with no security checks, nobody checking to see if you have 1 plastic bag with 3 oz. of liquids, etc. The train will beat driving because no one gives a train a speeding ticket for going 100 MPH.

A train is actually faster than flying.

Train: 3 hours 30 min ... get to Penn about 10 min before your train leaves, so you are at 3:40.

Fly: Get to JFK/LGA or EWR two hours before your one hour flight, get through security, fly, taxi, get to the gate, wait for your luggage, and get taxi into DC.

Ughhh.

You should find out if Scottie can beam you there?

If you want a serious answer, you need to give more information because obviously a flight would be quickest but that you already know, so what is it you need to base this on?

Quickest is plane and scheduled for one hour, although actual flying time is about 35 mins. Can also be done by train (about 3.5 hours) and car (about 3 hours if I remember).

Quick - fly, long and relaxing - train.
Or it's a long drive. Depends on how fast you need to get there and if you want to sight see on the way

It depends what you mean by the word 'best'. Do you want the cheapest or most scenic or cultural or quickest or slowest etc.
