A couple of candles to light by the bedside for a romantic night...
I would take a some medications like pain killers, cough medicine etc because the weather is not too good. Getting sick is the worst thing that can happen on an anniversary.

Toothpaste! I always seem to remember the toothbrushes, but not the toothpaste.
A roll of toilet paper - just in case they don't have any spare.
Unless you are having room service, it's a great idea to take some snacks and drinks. Motels only provide basic tea and coffee requirements, so taking some extras to eat or drink is a very sensible idea.

Toiletries, medications, contraceptives, enough socks/underwear, camera (you do plan on going outside at some point on your vacation???) cell phone chargers, bathing suits, a sweater or sweashirt.
I'm a chronic "Over-packer" so I normally have a lot of clean clothes after I travel and have a surplus of toiletries for traveling. Way to go me. LOL
