Seal a plastic bag around a branch, if you get condensation or water in the bag then you could say that the plant is transpiring, turning CO2 into O2 and H2O. These are the 2 byproducts of photosynthesis.

If there is light, there is photosynthesis going on probably (unless the plant is too stressed or damaged in some way). There are machines that measure photosynthesis, the company LiCOR makes several. They're pretty expensive and quite complicated to use.

Uh, what's the question?

Do you want to know how you can test if photosynthesis is occuring in leaves?

If that's the case, you can set up a device to measure oxygen, if the oxygen level increases and you've controlled all of your variables, you can assume that the oxygen is coming from the leaves.
