Unfortunately, you may not be able to avoid hurting her feelings. However, you can avoid making her feel worse by being honest and upfront NOW, and deliver your message in a kind and gentle manner. Perhaps pull her aside one day after work and say something like "I just wanted to make sure that you understand that I'm not ready for a relationship right now and what happened the other day was a one-time thing. I hope I haven't hurt you, but I just wanted to be honest and let you know where things stand." That way, you prevent any wondering and worrying on her part and get things settled and out in the open immediately.
And then, in the future, be careful of who you chose to sleep with and BEFORE you do it, consider the consequences of what will happen AFTER. Especially when it comes to fellow employees...you really have to be careful in regards to sexual harassment in those situations as well. good luck.

First and foremost: Don't S**T where you EAT! Even dogs know that...
Secondly--this is one of those situations that won't be friendly, no matter what you do. You screwed up, and you have to take the brunt. Either you man up and tell her the truth--that you think she's cool but you don't want a relationship with her--OR--you ignore her, flirt with other girls, act mean to her, so she'll leave you alone.
Basically, she's going to be hurt no matter what you do, and now the workplace is awkward for both of you.

don't tell her that you regret it for one. that hurts. but let her know that you're not looking for a relationship. that right now, it's date time in your life. that way it doesn't obligate you to get into a relationship, and you're still free to go out with anyone else, without destroying her feelings.

Tell her the truth. And don't be an A.S.S. by trying to flirt with any other co-workers. Just keep your personal life outside the workplace from now on. Never talk, admit, nor deny the brief encounter you had with her even if she does.

did you lead her on, if you did she has a right to want more from you and you can only apologise for being insincere, if you thought it was just casual sex then just go back to work as normal and if she persists then let her know she is a friend you value
