According to medhelp.org, Dr. Harvey Cushing (1869-1939), who was an American physician, surgeon and endocrinologist, first described a woman with signs and symptoms of this disease in 1912, and in 1932 he was able to link the adrenal overproduction of cortisol to an abnormality in the pituitary.

Harvey Cushing, a neurosurgeon. I think he discovered it in persons with a particular pituitary tumor. It was later described in persons who were administered large doses of ACTH and other steroids.
