First of all, what condition are you in now? how old are you? These are important.
Not exactly sure how far it is but three days isn't exactly pushing it. I would say try to start riding five days a week(taking off if you get too strained) for maybe an hour or two to start. you're going to have to get used to being on the bike for long periods. better gradual than all at once.
The thing I might suggest is making yourself push a little further each time you ride. It might hurt, bad, but fight through it. It'll develop the muscles you will need. Good luck and have fun!

I wouldn't say much training is necessary. I did a London - Amsterdam bike ride a couple of years ago and the only training I had was that I cycle to and from work everyday (an 11 mile round trip) and I found this to be plenty. Not sure how hilly the route will be but if you can find any big hills to train on it would be highly beneficial.

If it was a longer route a lot more training would be highly advisable (as I found when I went on a bike ride in China in October).

Depending on the route the whole trip could be less than 450 km so that is only 150km a day. If you ride as little as 20km or an hour (which ever comes last) a day you should be ready for it within a month. Of course the more time you spend on the bike the better.

After a week of one hour or 20 you will find that you actually want to go further

There are riders that would tackle this distance comfortably in one day. ( not professional)

If you're doing the organized ride, that's 300 miles in 5 days!!

Average 60 miles a day and that means LOTS of saddle time.

I'd ride until I was comfortable riding for 6 hours at a time, with a 10 minute break every hour.

Training in holding your breath for a couple of hours. I am sure that you are aware of the fact that London and Paris are seperated by a big splash of water.

A lot, But you don't even cycle a large proportion as you will be using a ferry or channel tunnel!!
