It means "the way" or "the path" Karate-Do translated literally means the "way of the open or empty hand" (with Kara meaning empty or open, and Te meaning Hand)
the way or path meaning also comes from the Chinese word Tao (actually pronounced Da-oh)

Do comes from the Chinese Dao or Tao. It has been variously translated as way, art & discipline. It can't be accurately translated. The best way to understand it is to read Lau Tzu's "Tao Te Ching". There are several English translations available.

yeah, it most litterally means "they way" or "art", and as an added piece of info to show my point tae-kwon-do can be broken down into tae (hand) kwon (foot) and do (the way, or art) thus, it means the art or way of the hand and foot. which is why it focuses more on foot techniqes than karate-do, which only has hand in its name.
