Most people will tell you the 30-06 is the 'best', but like most things in life, that's completely subjective. The '06 has killed countless deer in the past century, and it seems that most ammo in that size range is a one-off of the 30-06. Rounds like the 308, 300 family, 25-06, 30-378 Wby.....the list goes on and on, all have roots to the 30-06. So whats the perfect round? It depends on your hunting situation. If you are doing close range hunting, then a good ol' 30-30 would do just fine. Even a .243 Winchester would be great out to about 400 yards. Little rounds like a .222, .223, .204 Ruger, all have the potential to drop a deer. Many people might say the .308 Winchester is the perfect size. Others will go big and use a 300 WinMag and could drop a deer at ranges people don't even hunt at. It depends on how far you want to shoot the deer, and what kind of terrain you are in. To sum this up, get something that shoots a .30 cal bullet and you can't go wrong. BUT, there are always lots of options.

The best all around deer cartridge is the .25-06 Remington with the 115 to 120 grain quality hunting bullet.

Flat out, without qualifiers, it'd have to be the 7x57 Mauser or its ballistic twin, the 7mm08. The 6.5 and 7mm's traditionally are loaded with bullets that have good sectional density. You just don't hear about these things having problems setting up or penetrating poorly. Efficient and flat enough trajectories for almost any situation. The new premium bullets are very reliable, but traditionally, too little velocity means a rainbow trajectory and limited range, while too much means an occasional bullet will break up too soon. These cartridges suffer neither of these faults, and those who use them understand that the 7 Mauser's history ( since 1892!) of exceptional performance in the field was gained for good reasons. If you hunt with calibers smaller than 6.5mm long enough, you'll eventually find enough situations when it isn't enough gun, even in the overbore 25-06, and the 308 & 30-06 are really more than is needed for deer in the US.

I like the 7mm08. Short action which makes for a lighter gun and quicker bolt throw. Not a lot of recoil. Good choice of bullet weights. Accurate and it has enough energy to cleanly take deer at reasonable ranges. Of course this question will have just about every cartridge ever made given as an answer but that's what makes America great. Choices.

THERE IS NO SUCH THING.
There are SO MANY cartridges that are SO GOOD at dispatching whitetail that your question is literally impossible to answer.
Go with the .308, the most popular .30 caliber rifle, good for deer at any reasonable distance, BOATLOADS of ammo, and extremely accurate in a good gun. So good it's what virtually every purpose built sniper rifle is chambered in atleast in America.
If you want something a little different the .260 Remington would be my #2 for it's technically "superior" bullet.
But anything in the range from way up to way down from the .308 can be used.

The 30-30 has killed more deer than any other caliber in the U.S.A. Thats a fact!!!! I personally prefer the 30-06 because it is much more versatile with a greater variety of bullet weights! The famous Outdoor Writer Jack O'Connor swore it was the only caliber that a person ever needed and he used it in Africa on all the Wildlife that he hunted there including elephant, Rhino,Water Buffalo,Leopards, Lions!!! Need I say more??????? Happy Trails and Good Hunting!

If all I were going to do was hunt deer, I would choose one of the ultralight bolts in a short action in 7mm/08. A Remington model 7, or Ruger ultralite with a 20 inch barrel. This caliber will reach out to 250 or better, and still be more gentle on the shoulder when benching at the range. 140 grain bullets would be the best.
