In my experiences with weddings, the best man makes a toast to the groom and his new bride and shares a brief story about how knowing the groom has been good and why so and how this will make him an excellent husband and life mate for his beautiful new wife. Wish them many happy years to come and then bottoms up.

Speeches that are funny and end in how excited you are about all this are great. I gave a couple best man speeches and they all involved something funny my friend had done, and then I tied it into how excited we were about their new wife. My best man gave a great one about how my ideas always lead to...weird situations, but my best idea ever was happening right then. Funny, poignant, and concise...stick to that and you'll be great.

The tradition is to first embarrass the groom, then talk abut when he met the future-bride-to-be and about how much the groom was crazy over her,etc.
Then he wishes the happy couple well.
