a stopover is a break from the trip. say your coming from japan going to australia, you'll have your stopver in singapore for a few hours.from there you'll have to take another plane this time to your last destination which is australia.
it's a few hours interval. you can shop, dine,or rest at the airport lounge while waiting for you next flight.
the stopover is dictated by the planes you have. singapore airlines would have their stopovers in singapore, brunei airlines in brunei, malaysian air would be in malaysia..

1st : what explained by 'innerbeauty' & 'warrior is a child' is not stop over. they're explaining about transit.

2nd : stop-over is when you go to place A by flight which makes you do transit in place B first, and you are staying at place B for more than 24 hours. that's called stop-over.

real e.g :
1. I'm Indonesian and traveling to Japan by Garuda Airlines. I'm taking flight from Jakarta - Denpasar - Japan. I'm landing at Denpasar and I'm staying there for about 6 hours before I'm flying to Japan = transit
2. I'm Indonesian and traveling to Japan by Garuda Airlines. I'm taking flight from Jakarta - Denpasar - Japan. I'm landing at Denpasar and I'm staying there for more than 24 hours before I'm flying to Japan (just imagine that I'm flying to Japan on the next 2 days...) = stop-over

hope it's quite clear for you...
