When I use that expression, I mean absolutely yes. The key is the word "definitely." If someone says "most probably" they generally mean "probably"...the word "most" just underscores the second word. Actually, the phrase "most definitely" is sort of redundant, like when someone says something is a "true fact"...is there any other kind of fact?

Ninety nine percent of America does not care about standard English so I won't bother telling you that "definitely" cannot be qualified. Let's just call it hyperbole.

That is the definite truth. That is "most definitely" the most handsome man. It is such an every day phrase that it is hard to tell the meaning. How about the word "really?" That is "really" the most handsome man. Yea, what do you think?

Though not really grammatically correct, it means "certainly" or simply "definitely." The "most" is not needed, but it is used to add extra emphasis.

It means I assure you, or the best thing to do. Definitely means correct and most means the best.

"Most definately" is basically over emphasizing "definately".
