Surest way I know is to become his friend!
If you can't make peace, either avoid the person altogeather or avoid him when you are alone.
The short answer is: Given the situation (no weapon or background in self defense), you CAN'T defend yourself in 'hand to hand' so don't even think about it!
That's why they make pepper spray.

ok lets get real, a person twice your size has a great advantage in a stand up game. Time for a ground game...
Now try to lift a 300lb person, heavy huh.
Now take that same person and put them in a swimming pool, now you can lift the weight. That is similar to the ground and the effect you can have on manipulating their body weight, plus you can place them in positons taking away their weight advantage.
Remember always fight a single limb, joint, never the person. You can take your body weight and with proper leverage and body mechanics you can break a limb very easily regardless the size of an opponent. A broken hand can stop a large man fast.

you have a weapon.it's called your body.and almost every part of your body can be used in the right situation.
A good well rounded style would be freestyle Karate. You will learn stand up,ground, grappling and weapons. What a lot of people don't understand is that freestyle Karate is about practical street defence not who can kick the highest or punch a pad the hardest. It's definitely not a sport martial art like TKD or BJJ.The motto of my style is the best of everything in progression.Basically that means we don't care where the technique comes from we improve it and integrate it into our style while still maintaining tradition as do most freestyle Karate's.
The hardest thing is finding a good experienced instructor.I would recommend Bushi Kai or Zen Do Kai, but if your not in Australia or New Zealand you may have some difficulty finding some one who teaches these styles. These styles also usually have separate classes available to everyone in Muay Thai and BJJ/Submission/Shoot wrestling. If you can't find one of these i would suggest Kempo or Enshin or another freestyle Karate.
GROUND GAME-where do these idiots come up with this crap?with out training you will get slaughtered on the ground.the last place you want to be is on the ground in any situation unless you want to get your head kicked in.

Well now, the boot is a great weapon. Not the sneaker, the boot. Works really well applied with great force to the knee, or the shin, and even better if turned sideways, kick into his shin, then drag your foot down his shin and stomp his foot as if you were trying to kick start a Mack truck. Shove your thumbs into his eyes is good - the deeper in the better. Grab him by the boy parts, squeeze for all you're worth, and try to lift him off the floor is also good. Mike Tyson has had good results biting ears off.

stop focusing on his strengths, and look for weakness. if there isnt any, make some. no matter the size, strength or tolerance of pain, the person in front of us has the same weak spots as you.
knees, bones of the foot, throat, etc
you really do need some form of training to act as a foundation though, but the trick is to make cash in on the opponents weaknesses. you wouldnt fight Mike Tyson at boxing, or you die, so you need to improvise and go for the things that he will notice.

I assume it is too late to start Martial Arts lessons. Either use a weapon or run. I recommend the latter.

"The bigger they are, the harder they fall" I am 5'8" and have taken out guys twice my size by knowing where to strike. You don't want to fight with or go toe-to-toe with a guy that big. You want to strike areas where no matter how big, strong, or muscular the guy is these areas remain soft (eyes, filtrum, throat, groin, knees, etc). And as far as big guys go, most people are intimidated by big guys mainly because there are big. I've have many more challenging run-ins with guys who are more my size. "It ain't the size of the dog in the fight, it's the size of the fight in the dog"

Rent Karate Kid. That old geeser Pat Morita was able to whip a whole gang of big, tough high school kids - maybe his technique will work for you. I noticed that when he does spinning kicks, his leg never gets above waist-high.
Seriously, go for the knees and groin. (and then enter the witness relocation program)

avoid ending up on the ground, because you are inexperienced and he will just flatten you. stay standing, throw punch after punch and don't stop until you finish him or he finishes you. If he get close knee him in the balls or kick him in the balls. If you end up on the ground try to stay on top and choke him out. Remember fighting dirty is probably the smartest thing you could do in a fight like this.
