The brief searches we've done on the internet talk a lot about drug abuse and the like, which is not an issue here. It has become a hassle for her, in that rooms seem darker than they really are. What is most concerning is the idea that this may be a symptom of some bigger problem. Our family has had its share of weird and rare medical conditions, so something straight out of a "HOUSE" tv script would not come as too big of a surprise.

Your only hope for diagnosis would be to get your wife to an optomitrist to examine her eyes and her pupillary response to light. Further follow-up with an opthalmologist (an MD who specializes in eyes) would be needed if the problem cannot be diagnosed. Annual eye exams are important in order to look for signs of cataracts and glaucoma. You're probably due yourself if it's been more than 2 years since you saw one.

i would start with a visit to the eye doctor, proceed from there...
