Can be interpreted in three ways:

1. Can you be my date on Valentine's day?
2. Can you be my date forever?
3. I have nobody to hang out with on February 14.

It all depends on the context. When someone asks "Will you be my Valentine?", it's typically meant in one of two ways:
1) They want you to be their date on valentine's Day
2) They want you to be theirs - as in 'will you be mine?'. The second could be interpreted many ways though with 'will you be my valentine' being used in a marriage proposal, asking someone to be their boyfriend/girlfriend, or a partner or spouse reaffirming that they're committed to one another.

Sweet!! Did someone really ask you that?! I think it means someone wants to be your sweetheart. Have fun!
