No - I know of single mating that produced 10 pups and multiple ones that produced a single.
Save in a few rare cases where the males fertility is very marginal it's all on how many eggs the female ovulates. This is partly genetic (some breeds/family lines always prodce small litters) and a little her age and nutritional status

no it won't . I had a dog that was bred once whelp 13 puppies and then others were bred multiple times to ensure conception and they had a smaller litter.
If you do want to make sure she takes, breed her in 2 or 3 day intervalls.
