No. Well, yes, insofar as they are both birds, but the Atlantic Puffin is in the order of Charadriiformes, which includes waders, gulls, and auks. They live in the Northern hemisphere. Penguins are in the order of Sphenisciformes, which is strictly penguins, and they live in the southern hemisphere.
