Check out the discovery channel. They sometimes show a documentary where fighters from different styles are tested and measured using the same technology for crash test dummy's! It explains the amount of force and the angles required. Basically its when the brain suffers massive trauma from a blow that causes impact on the brain! The brain sits in its own case called the dura and is surrounded by fluid. This then sits inside the skull so a sudden movement can cause the brain to rapidly move from side to side.

Again its all down to nature if you are a big bloke then your chances of knocking someone out with one punch are greatly increased. That's just physics I'm afraid.

There always seems to be a fascination about knocking someone out and i think this has more to do with sports martial arts than traditional ones. Remember there's more than one way to skin a cat!

"There is actually very little known about what exactly causes a knockout but many agree it has to do with minor trauma to the brain stem. This usually happens when the head rotates sharply, often caused by a strike. A common misconception is that a knockout is caused by compression put on the carotid arteries in the neck restricting blood flow, but this is in fact how a choke out works."

Punch to the jaw jars the brain causing knockout. Other strikes can cause a sudden drop or spike in blood pressure. Chokes deprive the brain of oxygen.

In medical terms, there are three ways to cause unconsiousness as it relates to martial arts.

Concussion:

An injury to the brain – usually caused by a blow to the head. It is a temporary loss of consciousness and normally followed by rapid recovery.

Cerebral compression:

A condition of increased pressure inside the skull that compresses the brain tissue and disrupts brain function. It is caused by a significant blow to the head causing bleeding or the formation of a blood clot.

Skull fracture:

A break in the continuity of the skull bone

the main cause is your brain hitting the inside of your skull from a blow,a fall etc.the other major causes are loss of oxygen or blood to the brain.a knockout from a punch to the jaw is usually caused from your brain hitting the inside of your skull.

Usually the head is forced upwards and backwards by the blow resulting in a momentary interruption to the blood supply to the brain. This causes the person to pass out or become unconscious. If the blow is very hard further damage can be caused to the spinal column which can result in coma or even death.

movement inside the skull from an impact puts strain on the tiny nerve connections that carry electrical signals around your brain. when you take a big enough hit, the signals are temporarily interrupted and you lose consciousness.
