Ok that last answer was so lame I couldn't pass this question up!
My answer . . . probably just as lame.
Just do it!
Talk to people who are doing it already. Local radio DJ's, go in buy them lunch / a drink and ask them questions. If your cool they will be more than willing to answer your questions. Ask what would make a good demo.
Make a demo. I've checked in my area and you can get time in a professional recording studio for as little as $25 / hr.
So gather together your best material hit the studio and then hit the streets.
Send it to local radio stations, advertising agencies, and talent agents.
Remember it's not usually what you know but who you know. So, get out there and meet people who are doing what you want to do.
Good Luck to you!

If I were going to do this, I'd start by listening to ads or other works that were like the things I'd want to work on, then duplicate the voice with your own. You don't have to find a karaoke for voiceover, but just listen to others.
Then, sit down at your PC with your microphone, and using Windows (Mac??) Sound Recorder, you can practice making your own sounds and ads. When you have a good one, or several, burn them to a CD or email them to a radio station, recording studio, and/or TV station, and let them know you're interested in getting started.
Keep after it, it's a hard field to get into. Oh, and worship Don LaFontaine. He's the King of voiceover!
