You first see the Mirror of Twilight in the Gerudo Desert, at Arbiter's Grounds. To get to the desert you need to fly there via the cannon at Lake Hylia. And to get flight to desert, you need a letter from Auru, who is at the lookout tower, which you give to Fyer, who is in control of the cannon.
