National Express coach from London Victoria to Inverness leaves at about 11pm at night arrives about 11am the next morning. Takes about 12 1/2 hours. From there take a local bus to Wick, and then another local bus to John O'Groats. The latter might only be a summer service and very infrequent but should hopefully be doable before sunset on day two, by which time you'd have been travelling for the better part of 24 hours.

If you kept to local buses such as Stagecoach, you'd have to change countless times, doing each segment in small stages, pay far more, and probably end up sleeping overnight in several bus stations once services stop for the evening and you can't get any further.

I guess you'd probably have to take the A1/M1 corridor through the North East. Probably something like London - Watford, Watford - Luton, Luton - Bedford, Bedford-Huntingdon, Huntingdon-Peterborough, Peterborough - Grantham, Grantham - Newark, Newark-Worksop, Worksop-Sheffield, Sheffield - Barnsley, Barnsley-Wakefield, Wakefield - Leeds, Leeds - Harrogate, Harrogate - Ripon, Ripon-Northallerton, Northallerton-Darlington, Darlington-Durham, Durham-Newcastle, Newcastle-Morpeth, Morpeth - Alnwick, Alnwick - Berwick, Berwick - Edinburgh, Edinburgh - Stirling, Stirling - Inverness, Inverness-Wick and Wick-John O'Groats. One helluva journey, but don't quote me on the exact route !

Yes theoretically it is possible as long as you do not mind a little walk between bus stops etc. It would take ages and you would be really disappointed when you got there. I was gob smacked when I got there last October to discover that you had to pay for a guy to put up the sign post saying Lands End ??? miles. It is positively bleak!
Scotland hosts some truly awesome sights John 'O' Groats is not one of them.

It would certainly be possible although much easier in the summer peak. If you can get yourself to Inverness on one of the major bus lines, then from there citylink run to Wick. From Wick there are regular departures to John O groats by one of the local bus companies. Would take a while though, well over a day.

I don't know about the time it would take travelling the whole length on a bus, but I did once travel from Sevenoaks in Kent to Thurso and it took 23 hours on the train.
You could 'break it down' into time zones by finding out how long it takes by bus from city to city and adding up those times.
Have fun!
