There are actually lots of differences between Land Cruisers and the Sequoia. Here are some of the biggest differences:

1 - The Sequoia is sold only in the United States and Canada while the Land Cruiser is sold world-wide. This is probably the biggest difference as the Sequoia is specifically designed for the North American market.
2 - The Sequoia is based on the Tundra platform where the Land Cruiser is a unique frame/platform.
3 - From a price perspective, the Sequoia is slotted between the 4Runner and the Land Cruiser.
4 - The suspension systems are different from each other.
5 - Engines are different.
6 - There is considerably less aftermarket support for the Sequoia (accessories for either speed or off-road).
7 - The Sequoia is considered to be less capable off-road compared to the Land Cruiser. While this fact depends on what you consider off-road and the experience of the driver, it also has to do with the suspension and traction control systems.
8 - The Land Cruiser has a history and huge following which the Sequoia does not enjoy.

The Land Cruiser is a bit smaller than the Sequoia and is based on a completely different platform. I believe the engines are different as well. The Land Cruiser has a more sophisticated 4x4 system and is better designed for off-roading, whereas the Sequoia is just a family hauler with the capability to tackle the snowy mountains in the winter. Don't forget that the Land Cruiser's base price is a lot higher than Sequoia's and it is more luxurious than it's comparison.
