unlike air conditioning, running the heater does not increase fuel use.
So run the heater and wear what makes you a safe and comfortable driver.
I prefer the heater, and take the coat off as soon as the car is warm.

Taking your jacket off while driving is less restrictive, whether you crank the heat up is entirely a personal choice.

I'm always cold, so I wear my coat AND crank up the heat. I also like to put my gloves on the heating vent, then put them on right before I go outside for a small piece of warm-hands heaven.

Taking off your coat in the warm car gives you a chance to add a layer of insulation when going into the cold air. This makes it easier for your body to adjust to the large temperature difference. Helps you stay healthy.
It's February in New Hampshire, and the only sickness I have is I'm still riding the motorcycle in the picture. Yes, that's snow in the driveway.

You'll use less gas if you keep the jacket on & lay off the heat.
But really, it's personal preference.
