!. SYDNEY- absolutely beautiful city and lots to do as a tourist traveling alone - Sydney Explorer Bus (Hop on/Hop off) will keep you busy and let you see so much!!!... been there..done that and would do it again!
2. CAPE TOWN , South Africa - again, a lot to do, waterfront harbours & shopping, gorgeous! It's a tie between #1/#2. Fun for the adventurous types.
3. ZAMBIA-Victoria Falls - It's one of the 7 wonders of the World, which is what took me there. I was not disappointed. Elephant safaris, microlight, ziplines across the gorge...lots of adventure activities for those who like them.
4. RIO DeJaneiro - Christ Redeemer Statue.. fly a helicopter around it! So cool :) Beaches, music (beware of crime though....)
5. GREECE- Acropolis & Parthenon -island hopping-beaches!!
6. ROME - The coloseum was pretty cool- lots of history
7. ITALY- Florence was pretty, Venice & St. Marks square
8. PHILIPPINES- So many islands to choose from-great xchange rate! Your money will go really far there!
9.CAIRO, EGYPT- Cruise the nile and see the Sphinx
10.BEIJING, CHINA- The Great Wall of course

St Petersburg and Moscow, Russia - beautiful and unique architecture, great vodka, caviar, interesting culture

Prague, Ceck Rep. - Oldest and most preserved city in the world; just to see buildings build before WW I and WW II and still standing like they were built yesterday is fantastic.

The consentration camps in Poland - so many years ago and you get such a weird feeling looking and the buildings and the grounds, and experience you would not forget.

Lvov (the city of Lions), Ukraine - again great buildings and even more intresting on the inside of them, awesome cafes, great food (Cheep)

In all these cities people party way into the wee hours of the morning.
Have lost of fun!

I'm going to answer with one you can actually do in a RTW trip! Because of airline partnerships, etc., some of the suggestions made here are impossible to do in one trip.

I like Southern Hemisphere, and let me just surmise that you are starting from London (if I may):

Buenos Aires (Argentina) - Santiago (Chile) - Easter Island - Tahiti (French Polynesia) - Apia (Samoa) - Auckland (New Zealand) - Sydney (Australia) - Ayers Rock (Australia) - Perth (Australia) - Cape Town (South Africa) - back to London. These are all direct flights on a RTW circuit which you can actually do, and not just dream about.
