Penicillin is a very powerful antibiotic discovered in 1929 by Alexander Fleming. He noticed that a fungal colony had grown as a contaminant on an agar plate streaked with the bacterium Staphylococcus aureus, and that the bacterial colonies around the fungus were transparent, because their cells were lysing. The substance was named penicillin, because the fungal contaminant was identified as Penicillium notatum. Later it was found that penicillin was active on many different bacteria strains.

Chemically, Tpenicillin is not a single compound but a group of closely related compounds, all with the same basic ring-like structure (a beta-lactam) derived from two amino acids (valine and cysteine) via a tripeptide intermediate.

Many people were saved from previosuly considered deadly infections by penicillin (such as those due to Staphylococcus), since the 2nd World War when the antibiotic started being produced industrially.

"greatest" may be overstating it a bit...
It is important because it is the first antibiotic made from a cultured organism.
It is useful for treating certain bacterial infections.
However, its greatest impact will ultimately turn out to be negative, in that in a large population with repeated use, it effectively selects for bacterial that are resistant to antibiotics, thus allowing more virulent strains of disease to arise.
