You said it was a technical question, and since it's in the engineering area instead of word play, I'll assume you don't want the 'you won't find an elephant with one hand' answer.

The most common species of elephant, the African Bush Elephant has an average mass between 7000 and 10000 kg (15000-22000 Lb).

To lift something like that one-handed would require some sort of mechanical help. This could be a lever, but it would have to be a very long lever to multiply that amount of work, and at that mass, it would have to be very thick to keep from bending or breaking. A very long, very thick lever would not be possible to move into place one-handed, nor would you be able to lift it onto it's fulcrum one-handed (I'm keeping with the spirit of the question - if you can't manipulate the tools into position one-handed, then you aren't really doing the work one-handed). A better solution would be a chainfall, block and tackle, or some other type of hoist.

Of course, there are some simpler ways (non-technical, though) to do it (If you're a fan of lateral thinking problems, think of these as the 'Wally' answers): Train the elephant to recognize hand commands, put the elephant on a freight elevator, use a rope and guide him up some stairs, etc.

you'd need like a mechanical advantage of 400, assuming you can lift 50lbs with one hand and the elephant weighs 20000.

either that, or genetically engineer the elephant down to chihuahua size.

That replaced into stable- I enjoyed that. ok so an elephant lays an egg on the stairs- you may no longer push it up, you have no potential of flying it up or lifting it up. How do you get the egg up the stairs? answer: Elephants do no longer lay eggs! Hahahaa

It is called their trunk.
