i know some people in Malta whose surname is Spagnol.i don't know is something to do with Spain .

It sounds like the word, Espagnol, so maybe the original settler with that name was simply known as the Spaniard, being at the time the only one. Many names get worn away and abbreviated over time in this way, especially in times when literacy wasn't common: witness London pub names, The Elephant and Castle was originally L'infanta del Castille (the princess of Castile) and the Bag o'Nails was the Bacchanalia (from the drunken revels of the God Bacchus and his followers.)
