Not sure about vista yet, but there are numerous improvement in Office 2007 over Office 2003. Vista looks nice if you get one of the good versions (it comes in a few flavours / flavors).

Like all new OS, you are probably going to need a hardware upgrade as well to get maximum benefits.

The Interface (Aero Glass), The Security, a majority of spyware and viruses won't run on it (but you still need anti-virus and anti-spyware), you'll be doing fewer scans or worrying. The ease of use, how it seemingly and magically corrects hardware, software or driver conflicts without pestering you about this and that. All it does is ask permission to perform changes and in my case, it has yet to be wrong.

I've been using Vista for about five days now (got it early from Newegg), it makes XP look like that garbage that it truly is.
