This question has been asked at least 5 times in some form or another in the last week,alone. The answer, again, is the .500 S&W Magnum, for commercially manufactured pistol/cartridge combinations. The .44 Magnum and .454 Casull don't even come close.
There are others that utilize rifle ammo, or non-commercial cartridges, known as "wildcats", but many people don't count these.

Well there are some questions to your question (you gotta love it). First off if you are talking "Revolvers" the 44S&W round lost it title many years ago to the 454 Casull. At first the **** Casull round was a "WildCat Round", Handloaders only. Then as Americans we are always looking to make improvments, and along came other wildcats, but today you can now buy those 454 Casull rounds at your Hi-end Sporting goods supplier.
In revolvers the S&W 500 Magnum holds the foot-pounds in engery but, the Ruger 460 comes in at a very close second, but has a faster feet per-second. NOW ONTO THE 2nd PART.
Single shot: Thompson Center Contenders! These come in so many variations or combinations it's not funny (WOW!). One frame from 22 rimfire all the way up to rifle cartridges like the 444 Marlin. I myself own a Super 14 with two barrel options, 233 Rem. (5.56 NATO) or a 35 Rem.
Next up: WildCat single shots by JDJ or also known as...JD. Jones. Jones first started out with the Super 14 frame from T/C and placed his own barrels on it...WHA LAH! the Worlds most POWER HANDGUNS. These were 444 Marlin brass necked down to a specfic calibar vai,..264 JDJ, 7mm JDJ, 411 JDJ comes to mind (many others). the Powerful 375JDJ has taken every dangerous species on the African Continent.

The .500 S&W, to the best of my knowledge, is the most powerful handgun-specific cartridge in the world. But some posters here have been incorrect- rifle cartridges modified for handgun use (with a long enough barrel, that is) are more powerful than the .500 S&W.

There are a number of 'pistols' now chambered for rifle bullets including the .600 Nitro Express. Other than these, you have the Desert Eagle available in .50 Caliber Action Express. This is a 'true' pistol; there are revolvers chambered for a new wave of big bore calibers such as the .454 Casull (which has been around the longest). All are more powerful than the old .44 Magnum.

The s&w .500 magnum is now the most powerful handgun in production. It even more powerful than most of the riffle cartridges shot through a handgun.

Probably the .500 Smith & Wesson. I have a .454 casull and think it's a close second though.

smith & wesson say that referring to the .500 that they now make. as far as the most powerful, the most is the .825 that is gona come out soon. there was another question just like this, and someone gave a link to it. i can't remember the name, but goggle .825 handgun and you should find it.

44 mag is the most powerful production gun but now there are .500s and 454 and so on. I own 2 S&W M29s one with a 4'' and one with a 6 1/2'' just love them and believe me they are powerfull enough.

I think there is a 50 caliber that will knock you on your a ss, I own a 45 Ruger that pack a wallup.
