Requirements for dependents are listed toward the front of instructions for form 1040. No, a spouse is never a dependent. If you file a joint return though, you'll get an exemption for each of you. You can file a joint return even if only one of you has income.

If you file as married filing separately, then you each take your own exemption. If only one of you files, the exemption for the one who doesn't file is lost.

No, you cannot claim your spouse as a dependent.

You generally will pay the lowest tax if you file a joint return. You will get both of your personal exemptions on a joint return.

If you file separate returns, you'll each claim your own personal exemption. Filing separate returns will normally result in higher tax liability and smaller refunds so most people should NOT file separately.

If you are married, you should be filing as:
married, filing jointly or
married, filing separately

Either way, your wife is your spouse, not your dependent.
