The hottest spice is the chillie,there are any number of varieties of chillie grown in chillie growing areas of the world & the pungency of each type varies a lot,it is said that the smaller the chillie the 'hotter' or more pungent it can be.

Tepín (Capsicum annuum v. aviculare), the tiny round fruit 1/4" across, is only harvested from wild plants growing in the dry desert of Northern Mexico, much hotter than habanero or tabasco. Plants are late maturing, usually requiring 120 days for green fruit and 200 days from the setting out of plants to mature red ripe fruit.

The hottest material I've ever heard of is capsaicin, the chemical that makes hot peppers hot. In nature, this occurs in the highest concentrations in habenero peppers, some specimens of which can be up to 1% capsaicin (by mass, I think, or possibly volume).

I believe that would be the piquin pepper, a tiny little thing with something like 300,000 heat units compared to the jalapeno which is somewhere around 60,000... My numbers might be slightly off but it's definitley the piquin....Google piquin pepper...it has a couple of other names....
