Good question. I've always thought about that too. I'm assuming we are talking about the real heavy wrestlers (400 lbs +). I think one thing sumo wrestlers would lack though is stamina. They could of course train themselves for that, but then they would probably drop weight. I think stamina and mobility like you said are the key factors. I don't know how fast sumo wrestlers are but I'd be surprised if they could run 40 yds in 5 seconds like most NFL lineman average. The size of NFL players has gotten bigger and bigger over the years. Here's a stat up to the 2006 NFL season: "From 1920 to 1984, there were never more than eight players in any season who weighed 300 pounds or more. This year, there were 570 players who weighed 300 or more listed on 2006 NFL training camp rosters, nearly 20 percent of all players." So it will be interesting to see how big these NFL players actually get.
