Fifth Ave in the 60's is nice. So is Central Park South. There is a new building in Columbus Circle where someone recently bought a penthouse apartment for over $50 million.

It really depends on what you are looking for, and what kind of place you want to live in. All the cool hipsters want to live in the East Village/Lower East Side or Williamsburg, but I would never want to live there!

That being said, I'm sure a lot of the hipsters would tell you to avoid the Upper East/West sides!

It really depends on what you mean by "best". You can find extremely nice places in rapidly gentrifying Long Island City, Queens, with a gorgeous view of the midtown skyline for a small fraction of what you'd pay for a much smaller place in Manhattan with either no view or a view of Queens.
