We vacationed there two years ago. Things I would include on the "must" list:

Statue of Liberty/Ellis Island.
Empire State Building
Times Square - Do it after dark for maximum effect - It was safe enough for us, at least before 10PM. Not sure about late-night though.
Ground Zero, although I'm not sure what status it is. It was not yet a new construction site when we visited it, and was very solemn - also the church across the street next to the Millinium Hilton was open as a "museum" of sorts honoring the rescue workers who dug through the rubble.
We used the subways to get around instead of cabs. It was actually a neat way to experience the city, and was safer then I was afraid it would be.
We took a tour on the red double decker tourist buses. The one we did covered Brooklyn via the brooklyn bridge, as well as harlem and many other parts we probably wouldn't have seen otherwise. Ride on the upper open deck.
Spend a day on canal street in chinatown, and buy a fake rolex or designer purse. but be sure to barter, they will drop the price to a third or better what they have it tagged for.
While in Chinatown, eat at a "Dim-Sum" chinese restaurant.
There is a pizza place across the street from where you get off of the Liberty/Ellis ferry. I wish I could remember the name, but it had GREAT NY style pizza.

Be sure to make a reservation for the pretheater dinner special at the Four Seasons, which is a five star restaurant and very romantic. If you are seated by 6:15 you get a full dinner from appetizer to dessert for $65 a person. Call 212-754-9494 and ask to be seated in the pool room. The best bargain in town but jacket and tie required. Get your theater tickets at the half price ticket booth or you can find discounts on line for a lot of Broadway shows.
Ride the Staten Island ferry, go shopping at Century 21 (across the street from the WTC). It's discount and very stylish. Metropolitan Museum of Art has everything-art, knights in armor, mummies, a small Egyptian temple and a nice place to have lunch. It's a full day experience. http://www.metmuseum.org/
Eat dim sum in Chinatown for brunch. Go to the Bronx Zoo. It's by donation on Wednesdays, other days it's $14

I am from NYC and it is too easy to answer this question. Definitely visit Times Square. If you like that kind of stuff like broadway shows then definitely go to Mama Mia! or Chicago. They are awesome! When it comes to entertainment then lounges in East village or Greenwich are free to get in and all you do is get yourself a drink and get yourself new friends. When it comes to restaurants then i suggest TGI Fridays and Olive Garden. Yankee game would definitely be something to remember. Just dont get lost trying all these things. Get maps beforehand so you dont look like a tourist. Not trying to scare you but it is very visible and not too safe. Most of the areas in the city are very user friendly. Oh and if you do get a cab then make sure you say this:"No short cuts and funny road please. I know where i am going." Otherwise you are going to pay over $30 bucks for something that would cost only $7-10 because he took you on a long run that is just 5 block away. ;) Good luck and have fun!

Visit the Statue of Liberty, (great view from the ferry- take popcorn or chips to feed the birds- Don't get ripped from the vendors there!)
Go see a show- On or Off Broadway!
Spend a day in Central Park, visit Times Square, Visit Times Square at night...
Maybe ask for a local's opinion while you are there for a "spontaneous" place to eat- Not necessarily a tourist trap or fast food...
There was a kool cafeteria...the AUTOMAT! Really neat place!

Curious George that was a terrible comment!

Just have fun in New York. Right now it's cold so I hope you can handle cold whether. I'm in New Jersey so I feel what New Yorkers feel as far as temperature is concerned. But you can go visit Madison Square Garden or Go to Time Square even go see a couple of shows. And don't forget to try the cuisine in New York because the food is fabulous! You have so many choices to choose from.

Well I hope you enjoy yourself. Have fun!

Must sees -- Central Park, Washington Square, Greenwich Village, Fifth Ave from 42nd to 59th (includes Rockefeller Center)other than that depends on what you like...Staten Island Ferry is Free and you get great views of the Harbor, and Downtown (Financial District) is pretty interesting as is a walk across the Brooklyn Bridge.

As for restaurants, that depends on your budget and taste you can get any kind of food imaginable and spend from $5 to $500 for a meal.

THe weather could be fantastic - 80 degrees blue skies no humidity or it could be hot and humid.

I grew up in NY and only recently moved out. I really miss the museums. The Met is my favorite, but the Museum of Modern Art and the Natural History are great too. It's kind of fun to see the big FAO Schwarz toy store on 5th ave (at 58th), near Central Park. You can try eating at Ellen's Stardust Diner, on Broadway at 51st? I believe. It's a 50's style diner with singing wait staff and great milkshakes.
