First you draft up a proposed 3D layout of the truss members that seems reasonably stiff (i.e., making sure that structure is neither indeterminate nor liable to linkage movement). Then you identify and locate likely vector forces upon the space truss (e.g., estimated roof loads distributed on each vertex). Then you matrix techniques for solving for the resultant forces on each member of the space truss. Using this information, you find the proper materials for each of the members (e.g., tubing of certain diamaters & thickness as well as type of steel). Once you have all this information, you go back to the beginning and do it again, but this time including the weight of the members. After having done that, do some flexure analysis and see if the structure flexs within acceptable limits under loading. Particular attention has to be paid to the possible bending moments on the vertex fasteners, which is a central design feature in space trusses. And so on. Reiterate as often as necessary until everybody is happy.

Same as it is for the earth. Lighter materials are used since we have to take them up there in a shuttle, but many of the same analysis used still includes gravity. Why? Because that same structure may have higher stresses of coliding vehicles/micro-meteorites/etc. Metal alloys are used primarily, as it provides both the tensile and compressive strength, and can withstand torsion predictably.
