Buddy, there's only one way to know for certain if a girl/woman is a virgin...that's if her hymen is still intact. And if it is, I highly doubt the gal in question will be letting you take a peek so you can know for sure. The best thing to do is get a book on female anatomy (or google it) and see what an intact hymen looks like. Otherwise, you're just going to have to ask the gal in question and take her word for it.
P.S. And just because a gal's hymen *isn't* intact doesn't mean she isn't a virgin. It could have been broken by other means, such as using tampons or having suffered a trauma of the vulva region (like slipping off a bicycle seat and straddling the front bar, hitting it very hard).
