the biggest change was the destruction of the Temple and the Diaspora. The religion changed from a religion of native people on their land to one in exile. Because of the exile, Judaism had to change it's focus from the temple system of sacrifice to one of personal observance of the law.

Reformed Judaism is probably the most changed. Other branches of Judaism (Orthodox and/or Conservative) have changed much less.
