She has the run of the house usually. I just started doing Pilates which requires alot of floor movement with various straps and balls. While I'm doing it, she will not leave me the f** alone. I know she wants to play. Is it wrong to lock her in the laundry room (litterbox is there) for 45 minutes while I exercise? I just feel bad becuz I've never done that before and to do it while she is in heat seems mean...

To everyone who said get kitty spayed---DUH. My vet (who I adore) told me it's better to wait until she is a year or so old, partly because she was abused as a kitten and I rescued her and he thought she should have plenty of time to adapt and to grow. She never goes outside--doesn't even try. Spaying is in her near future. Thx for the friendly advice re: free places. I can afford to do it. But more thx for the reminder that it's time.
As for "Sandwich"--your response was completely uncalled for. I never said it was a discipline problem. I put her in there so I can exercise and I don't have to keep shooing her away. Save your hostility for someone else, kiddo.

No not at all. If she has a litterbox, give her some water in there and 45 minutes is no big deal. It's not neglect as long as you still play with her at other times.

Yes it's bad!
I can't believe you're calling this a discipline issue! Your cat is victim of a terribly strong instinct she can't control. It's not like she is having fun being frustrated and suffering.
Not only she is being annoying, she is also suffering because what you think is playing, is in fact calling desperately for a mate who will never arrive, because she is indoors, and even if he does arrive, he will probably be ill and infect her and kittens. (only strays are not spayed now). SPAY YOUR CAT ASAP!
It's not her fault she's annying you, it's YOUR fault!!
If you want to keep a pet you have to have them spayed, otherwise you are the cause if she becomes annoying, and pines for weeks, in frustration.
So, have her spayed, and she will NEVER annoy you or anyone else again. AND, she will not suffer for days while she calls for a mate for nothing.
There are free spaying clinics, and if you haven't got enough money for the operation at your vets you can contact your local animal shelter for info on free or subsidized neutering.

We lock our cats in the bathroom when we bring things up the stairs because they always try to get out. I don't think it's mean at all.

She should be fine. If you give her a lot of attention before hand, and then again afterwards she probably won't even notice being left alone.
