A score is twenty or a set of twenty units, so five score is a hundred. The word "score" was used until quite recent times to mean twenty; we still say "scores of (for instance) people" to indicate a large but not enormous number.
It has Biblical connotations, as in the psalm "The days of our age are threescore and ten" - in other words a natural life span is 70 years.
Of course the word has several other meanings too, such as number of points achieved in a competition (football scores etc) and it can also refer to a kind of grudge or wish for revenge, eg to have a score to settle with someone. Whether this is linked with the old meaning of score as a unit of 20 is not certain.
