At work a co-worker I don't know so well lost his brother. Work is now passing around a card for us to write in. I've never experienced a death of a loved one, and wonder what should I write? I don't want to sound cliche or tacky. Please help.

Don't be afraid to sound "cliche". Conventional, familiar expressions are the most comforting. Misguided originality is what leads to tackiness.

Good question. It can be really difficult coming up with the right words.
I would suggest you just write something like
"so sorry to hear of your sad loss. Thinking of you.".
Then sign your name.
Something short and simple. If the person was a close friend you would be expected to write something longer.

All you can do is offer your heart felt sympathy and offer to pray for him/her during this time of grief.

Good luck!
