Open your photograph in Photoshop and copy it to make a new layer.

Make the top layer black and white (e.g. by using desaturate, or channel mixers) then carefully use the eraser to remove the top layer to reveal the coloured layer below. You could use a layer mask instead to stop it being a destructive edit.

I've done it here:

There are several ways... This is how I would do it...

Right-click on the photos Layer, and select Blending Options. Add a Color Overlay, choose white with a "Color" blend mode. (This is just making it Black and White...without losing the original colour.)

Add a new layer. Choose the Brush Tool. Make it the right size for the iris, and set the colour to whatever colour the eyes are. (You'll probably want to use a deeper colour then in original photo.) Then put the dots over the iris' of the the eyes. Use the eraser tool to trim off the top of the dot, and bottom if needed, so it fits in the eye. (Or if you have a stylus and tablet, you can easily paint in the iris.)

Now Right-click on the new layer, and change the Blending Options. Under the Blend Mode, change it to color. You may also want to change the %, or Blend Mode until it looks how you want... and even add a soft glow or something.

That's it.

create a path around the eyes (the coloured bit and black bit only) and copy them to a new layer.
on the main layer underneath, simply desaturate the layer and maybe have a mess with the levels, brightness etc to make it look nicer.

Select the eyes using whatever tool you want(lasso is good). Then invert the selection. Go to Image >Mode >Grayscale.
