Flying - less than an hour
By sea - a day or so
Driving - about 8 hours (watch out for those speed traps!)
Cycling - about 8 days (if you're fit)
Walking - about a month
Take your pick!

It's between 780 - 800kms. I live in George on the road to Port Elizabeth. I'm about 430 from Cape Town and about 350 from Port Elizabeth. So my estimate is accurate! But you can do as suggested by the lady above - go to:

NB: I disagree that the distance between CT and PE is 650. I lived in Plett before coming to George. More than 100% sure that distance between Plett and PE is not less than 250kms. I'm also 100% that the distance between Plett and CT is not less than 500km. All drive sites give the distance over 750.

The fastest is by air,there are number of carriers flying out of cape town to port elizabeth.Prices vary from around 40 to 120 pound for a one way ticket and flying time is about 1h:10m

About 10 hours if you drive quite fast, or if you poodle and enjoy the trip then as long as you like. Remember to use the hard shoulder to let faster traffic pass and give a quick flash of your hazards to say thankyou if they pull in for you to pass and a quick flash if oncoming move over for you. It would be a shame not to enjoy this part of the garden route, so take your time if you can. Happy tripping.

I have to agree with Papu , take his advice. I have driven the Garden route many times. It is a Stunning route ,however try to avoid doing this over Christmas and July holidays as roads can be packed in certain areas of eastern Cape. If you must , take a lot of of patience with you.

the thing is I know you said you just needed to get there most direct and fastest.
Well that would be flying - don't do that.
Drive during the day and plan to stay over. Just take the time out to break your journey. Perhaps in Wilderness or Knysna. If it is the only time you are ever going to be there, you will never regret doing that.
It is truly a special place to be.

If you can, plan to travle this route over 2 days - It is some of the MOST beautifull in the country. (I agree with the estimates from others. Shellgeostar should also assist with directions if required)

About 8 or 9 hours with one short stop driving but you can fly it in an hour or so which is quicker but the drive is very scenic

Fast and easy: flying.
It takes app 20 minutes from CT city to CT airport depending on traffic.
You must check in 1½ hour before flight (south african airways).
It takes 1 hour and 10 min to fly from CT airport to PE airport, plus driving to and from airports.
App 10 min to PE city.
So app 3 hours from CT city to PE city.

NATURE IS AT ITS BEST IN SPRING, EARLY SUMMER AND EARLY WINTER.DRIVING LIMIT IS 120 KM PER HOUR BUT YOU MAY DRIVE AT 140 KM PER HOUR AND IT WILL TAKE 6 TO 7 HOURS TO COMPLETE THE JOURNEY. YOU MAY FLY AS WELL AND FINISH THE JOURNEY IN LESS THAN 2 HOURS BUT MY FRIEND YOU ARE GOING TO MISS THE AMAZING BEAUTY OF THE GARDEN ROUTE. ONE LAST THING DRIVE ON YOUR LEFT HAND. ROAD IS QUITE WIDE AND TRAFFIC IS MUCH LESS THAN NORTH AMERICAN AND EUROPEAN STANDARD. YOU WONT REGRET IT.
