I think it largely depends upon your own sense of humor, but my personal favorites are The 40 Year Old Virgin and The Big Lebowski.
Another one that ranks high is There's Something About Mary. I saw it at the theatre for an early sneak preview, and it is the only time where the laughter in the audience was louder than the movie. For me, the movie was more wrapped up in the whole experience.

Anchorman hands down..ive never seen a movie that randomly funny or quotable with the combination...i love it and still quote it to today!! talladega nights and super troopers, grandmas boy follow...all hilarous movies.
