Spain was the most ruthlessly devout country in europe. Since they were also the richest country in europe at the time (new world gold) they were also the champions of church.
Protestentism had reached critical mass and the catholic church ruthlessly introduced countermeasures to implement policies of the reformation. The reintroduction of the inquisition was simply one mechanism.
Spain had only recently thrown the muslims out of spain. The muslims were much more tolerant then the christians so many jews lived in the lands the muslims had occupied. Spain ruthlessly christianized the muslims and jews that remained behind in the occupied territory.

Where else would you expect the Spanish Inquisition to be? If you're going to throw an Inquisition and name it after your own country, you might as well go whole hog at it.
